Tesamorelin occupies a narrow position among agents that act on the growth hormone axis. Unlike growth hormone itself, which is given as replacement, it stimulates the pituitary to release the hormone in pulses, so the downstream increase in insulin-like growth factor 1 depends on intact somatotroph function. Other peptides in the same family include shorter GHRH fragments and synthetic secretagogues with different stability profiles. Several points remain unresolved, including whether the reduction in visceral fat translates into fewer cardiovascular events, what happens to metabolic markers after long-term use, and how the drug compares with lifestyle or surgical approaches.
Tesamorelin is a synthetic peptide that belongs to the growth hormone-releasing hormone family and contains the same forty-four amino acid sequence as endogenous GHRH, extended at the amino terminus by a trans-3-hexenoyl group. That small fatty acid modification protects the peptide from rapid cleavage by dipeptidyl peptidase-4, the enzyme that shortens the half-life of native GHRH to only a few minutes. Chemically the compound is produced by solid-phase peptide synthesis, purified by chromatography, and supplied as a sterile lyophilized powder for reconstitution.
Regulatory approval in the United States came in 2010, when the Food and Drug Administration cleared the peptide for the reduction of excess abdominal fat in adults with HIV infection and associated lipodystrophy. The decision rested mainly on two randomized phase 3 trials that enrolled roughly eight hundred patients and ran for twenty-six weeks. Participants receiving active drug showed substantially greater declines in visceral adipose tissue than those receiving placebo, while total body weight changed comparatively little. A reformulated presentation was later approved, and the product has remained a niche therapy rather than a general weight-loss agent.
Measuring the effect of a growth hormone-releasing hormone analogue requires markers that reflect pituitary output rather than the peptide itself. The two most frequently used are growth hormone and insulin-like growth factor 1. Growth hormone fluctuates sharply across the day and responds to sleep, stress, and meals, so isolated readings can be difficult to interpret. Insulin-like growth factor 1 changes more slowly and is often treated as the more stable integrated marker of axis activity.
Because growth hormone is released in pulses, single measurements can misrepresent overall secretion. Investigators sometimes use repeated sampling or overnight profiles to capture the pattern rather than a single value. Provocative testing, in which a stimulus is given and the response is tracked over time, offers another way to characterize the axis. Each approach carries trade-offs between sensitivity, burden on the participant, and the influence of non-target variables.

Clinical study of tesamorelin has centered on adults with HIV-associated lipodystrophy, a condition in which abdominal fat accumulates while peripheral fat is lost. In controlled trials, treated participants showed reductions in visceral adipose tissue measured by imaging, alongside modest shifts in some lipid values. Effects on subcutaneous fat were smaller and less consistent across studies. Whether these changes translate into fewer cardiovascular events remains an open question, because the trials were not designed or powered to answer it.
Tesamorelin is a synthetic peptide that acts as an analog of growth hormone-releasing hormone, a natural hypothalamic signal. Its sequence corresponds to the forty-four amino acid form of the human hormone, with a small acyl group attached near the amino terminus. That modification slows enzymatic breakdown and extends the time the peptide remains active in circulation. The compound was developed as a pharmacological way to raise endogenous growth hormone output rather than supplying the hormone directly.
Stimulated growth hormone release leads to hepatic production of insulin-like growth factor 1, a key mediator of many growth hormone effects. In clinical studies, tesamorelin increased IGF-1 levels in a dose-dependent manner, although the response varies among individuals. The drug's effect on visceral fat is thought to involve growth hormone-mediated lipolysis and altered adipocyte metabolism. Muscle mass and lean body mass have also been assessed as secondary outcomes, but changes are generally smaller and less consistent than fat reductions.
Pharmacodynamic studies show that tesamorelin reduces visceral adipose tissue more than subcutaneous adipose tissue in the studied population. This selectivity may relate to differences in blood flow and hormone sensitivity between fat depots. Effects on glucose metabolism and insulin sensitivity have been investigated, with some trials reporting modest changes and others showing stability. The precise relationship between growth hormone exposure, IGF-1 levels, and visceral fat loss remains an active area of analysis.
=== Rebranding === On 24 September 2020, Giant Singapore announced that it would be undertaking a rebranding exercise, introducing a new logo and new features to its stores. It will also lower prices of daily essentials by 20% average for six months, following NTUC FairPrice's decision to lower prices in order to support Singaporeans during the COVID-19 pandemic. The rebranding of Giant's logo was extended to Malaysia in April 2021. As part of transformation, Giant introduces a section known as “Ringgit Zone” which has priced its products fixed at RM3 per item for bargain hunters. Both ShopSmart & G-Ekspres are being rebranded as Giant Mini.
=== Second Amendment rights === Gallrein said that "the Second Amendment is under constant attack from the radical left" and believes that U.S. citizens should maintain the right to keep and bear arms.
Hepatocyte growth factor (HGF) or scatter factor (SF) is a paracrine cellular growth, motility and morphogenic factor. It is secreted by mesenchymal cells and targets and acts primarily upon epithelial cells and endothelial cells, but also acts on haemopoietic progenitor cells and T cells. It has been shown to have a major role in embryonic organ development, specifically in myogenesis, in adult organ regeneration, and in wound healing.

However, this can be possible: for example, inhibitors based on the compounds mycophenolic acid and tiazofurin inhibit IMP dehydrogenase at the NAD+ binding site. Because of the importance of this enzyme in purine metabolism, these compounds may be useful as anti-cancer, anti-viral, or immunosuppressive drugs. Other drugs are not enzyme inhibitors, but instead activate enzymes involved in NAD+ metabolism. Sirtuins are a particularly interesting target for such drugs, since activation of these NAD-dependent deacetylases extends lifespan in some animal models. Compounds such as resveratrol increase the activity of these enzymes, which may be important in their ability to delay aging in both vertebrate, and invertebrate model organisms. In one experiment, mice given NAD for one week had improved nuclear-mitochrondrial communication. Because of the differences in the metabolic pathways of NAD+ biosynthesis between organisms, such as between bacteria and humans, this area of metabolism is a promising area for the development of new antibiotics. For example, the enzyme nicotinamidase, which converts nicotinamide to nicotinic acid, is a target for drug design because this enzyme is absent in humans but present in yeast and bacteria. In bacteriology, NAD, sometimes referred to factor V, is used as a supplement to culture media for some fastidious bacteria. High-cost unlicensed infusions of NAD+ have been claimed in the UK to be "clinically proven" and "effective" treatment for alcoholism and drug abuse.

== Oxidants and reductants == Oxidizing agents are added to flour to help with gluten development. They may or may not also act as bleaching agents. Originally flour was naturally aged through exposure to the atmosphere. Oxidizing agents primarily affect sulfur-containing amino acids, ultimately helping to form disulfide bridges between the gluten molecules. The addition of these agents to flour will create a stronger dough. Dehydroascorbic acid and potassium bromate are oxidants, acting on sulfhydryl groups and disulfide bonds in wheat dough, in particular oxidizing glutathione. Potassium bromate acts more directly or with fewer chemical conversion steps than ascorbic acid. Glutathione increases wheat dough's extensibility, or relaxes it, while oxidizing a dough's glutathione increases elasticity. Common oxidizing agents are:
=== Interactions with H3K4me3 and H3K14ac === The isomerization of the peptide bond between histone H3's alanine 15 and proline 16 is affected by the acetylation at K14 and can control the methylation states of K4. K4me3 represses gene transcription and depends upon the Set1 methyltransferase complex subunit Spp1 being balanced with the Jhd2 demethylases for proper function. Acetylation of K14 allows for a state change in P16 and primarily promotes the trans state of P16. This trans isomer of P16 reduces K4 methylation, which results in transcription repression. Isomerization of P16 has downstream effects of controlling protein binding to acetylated K18. When P16 is in the trans conformation, Spt7 is allowed to bind to K18ac, increasing transcription.

=== Gun laws === Sanders supports closing the "gun show loophole", banning assault weapons, and passing and enforcing universal federal background checks for gun purchases. In 1990, his bid to become a US Representative benefitted from the National Rifle Association of America opposing the competing campaign of Peter Smith, who had reversed his stance on firearm restrictions, and waiting periods for handgun purchases. In 1993, while a US representative, he voted against the Brady Handgun Violence Prevention Act (which established background checks and wait periods), and in 2005 voted for legislation that gave gun manufacturers legal immunity against claims of negligence, but as of 2016 he has since said that he would support repealing that law. In 1996, he voted against additional funding to the Centers for Disease Control and Prevention for research on issues related to firearms, but in 2016, he called for an increase in CDC funding for the study of gun violence.
== Applications == Deoxycholic acid has been used since its discovery in various fields of human medicine. In the human body deoxycholic acid is used in the emulsification of fats for absorption in the intestine. It has, in some countries (including Switzerland) been licensed as an emulsifier in food industry, but it is no longer common. Outside the body it is used in experimental basis of cholagogues and is also in use to prevent and dissolve gallstones. In research deoxycholic acid is used as a mild detergent for the isolation of membrane associated proteins. The critical micelle concentration for deoxycholic acid is approximately 2.4–4 mM. Sodium deoxycholate, the sodium salt of deoxycholic acid, is often used as a biological detergent to lyse cells and solubilise cellular and membrane components. Sodium deoxycholate mixed with phosphatidylcholine, is used in mesotherapy injections to produce lipolysis, and has been used as an alternative to surgical excision in the treatment of lipomas. Deoxycholates and bile acid derivatives in general are actively being studied as structures for incorporation in nanotechnology. They also have found application in microlithography as photoresistant components. In the United States, deoxycholic acid, under the brand name Kybella, is approved by the Food and Drug Administration for reducing moderate-to-severe fat below the chin. When injected into submental fat, deoxycholic acid helps destroy (adipocytes) fat cells, which are metabolized by the body over the course of several months. Kybella is produced by Kythera Biopharmaceuticals.
=== Thermoresponsivity in water === Polymer solutions that show thermoresponsivity in water are especially important since water as a solvent is cheap, safe and biologically relevant. Current research efforts focus on water-based applications like drug delivery systems, tissue engineering, bioseparation (see the section Applications). Numerous polymers with LCST in water are known. The most studied polymer is poly(N-isopropylacrylamide). Further examples are poly[2-(dimethylamino)ethyl methacrylate] (pDMAEMA) hydroxypropylcellulose, poly(vinylcaprolactam), poly-2-isopropyl-2-oxazoline and polyvinyl methyl ether. Some industrially relevant polymers show LCST as well as UCST behavior whereas the UCST is found outside the 0-to-100 °C region and can only be observed under extreme experimental conditions. Examples are polyethylene oxide, polyvinylmethylether and polyhydroxyethylmethacrylate. There are also polymers that exhibit UCST behavior between 0 and 100 °C. However, there are large differences concerning the ionic strength at which UCST behavior is detected. Some zwitterionic polymers show UCST behavior in pure water and also in salt-containing water or even at higher salt concentration. By contrast, polyacrylic acid displays UCST behavior solely at high ionic strength. Examples for polymer that show UCST behavior in pure water as well as under physiological conditions are poly(N-acryloylglycinamide), ureido-functionalized polymers, copolymers from N-vinylimidazole and 1-vinyl-2-(hydroxylmethyl)imidazole or copolymers from acrylamide and acrylonitrile.
Acetals, as already pointed out, are stable tetrahedral intermediates so they can be used as protective groups in organic synthesis. Acetals are stable under basic conditions, so they can be used to protect ketones from a base. The acetal group is hydrolyzed under acidic conditions. An example with a dioxolane protecting group is given below.

It is a laboratory-made peptide of forty-four amino acids whose sequence matches human growth hormone-releasing hormone, with a modified amino terminus. The modification is a short unsaturated fatty acid chain attached to the first residue. This change slows enzymatic breakdown and lengthens the time the peptide stays active in circulation.
The clinical program was designed around HIV-associated lipodystrophy, a condition in which fat accumulates abnormally around the internal organs. Trials enrolled that specific population, so the evidence base covers it rather than the general population. Regulators approved the drug for the studied indication only, and promotion outside it is not permitted.
Growth hormone therapy supplies the finished hormone directly, while this peptide acts upstream and asks the pituitary to secrete its own. That difference means the response depends on a functioning pituitary and on the body's normal feedback loops. It also means the circulating hormone profile is pulsatile rather than a flat, injected level.
It varies slowly and reflects cumulative axis activity rather than momentary secretion. Growth hormone is released in pulses affected by sleep, stress, and meals, making single readings hard to interpret. The slower marker gives a more stable picture across a study period.
