Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
A documented effect of tesamorelin is a reduction in visceral adipose tissue in some study populations. Researchers have reported decreases in trunk fat measured by computed tomography alongside changes in lipid markers. The mechanism is thought to involve growth hormone-mediated lipolysis, though the precise contribution of direct versus indirect pathways is not fully resolved. Studies have generally examined defined groups over finite periods, so long-term outcomes are less well characterized. Findings have not been uniform across all trials.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone (GHRH). Its sequence corresponds to the 44-amino-acid form of human GHRH with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification slows enzymatic cleavage and extends the peptide's activity relative to the native hormone. The compound is produced by solid-phase peptide synthesis and supplied as a lyophilized powder. Researchers classify it as a GHRH receptor agonist. Its structure places it in the same family as other growth hormone secretagogues that act on the pituitary.

Receptor-level activity begins when the peptide binds the GHRH receptor, a class B G-protein-coupled receptor found on pituitary somatotroph cells. Occupancy triggers Gs-mediated activation of adenylyl cyclase and a rise in intracellular cyclic AMP, which in turn promotes synthesis and pulsatile release of growth hormone. Because the compound acts upstream of the growth hormone axis rather than supplying hormone directly, its effect depends on intact pituitary function. Binding studies in cell culture and animal models have established this pathway; the detailed kinetics of receptor recycling in humans remain less well characterized.
Physicochemical behavior is dominated by the peptide backbone. The molecule is hydrophilic and carries a net positive charge near neutral pH, owing to several arginine and lysine residues. In solution it adopts a largely unstructured conformation, and aggregation is a known concern for peptide products of this size. Oxidation of methionine and deamidation of asparagine or glutamine residues are the principal chemical degradation routes. These liabilities shape how the material is formulated, handled, and analyzed, and they explain why lyophilized presentations are common in research settings.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, built from 44 amino acids. Its sequence follows the natural human GHRH(1-44) backbone, with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification blocks recognition by dipeptidyl peptidase IV, the enzyme that rapidly truncates the native hormone in circulation. The result is a molecule with a substantially longer plasma residence time than unmodified GHRH, which makes it practical for clinical and laboratory study.
Fifth generation In the 1990s, the fifth generation of silicone-gel breast prosthesis contained a semi-solid silicone-gel with a viscosity that reduced the occurrence of filler leakage and the occurrence of the filler migration throughout the woman's body of any silicone-filler that leaked from the implant-pocket. The plastic surgery studies Experience with Anatomical, Soft Cohesive Silicone-gel Prosthesis in Cosmetic and Reconstructive Breast Implant Surgery (2004) and Cohesive Silicone-gel Breast Implants in Aesthetic and Reconstructive Breast Surgery (2005) reported lower incidence-rates of capsular contracture and lower incidence-rates of shell rupture, and rates of medical safety and technical efficacy greater than the rates of safety and efficacy of early-generation prosthetic breasts.
A sickle cell crisis occurs when red blood cells switch from the normal saucer-like shape to a sickle-like shape, which can obstruct small blood vessels; an attack can be set off by temperature changes, stress, dehydration, and high altitude. A person with a single abnormal gene does not usually have symptoms and is said to have sickle cell trait; these people are also referred to as carriers. Diagnosis is by a blood test. Some countries test all babies at birth for the disease. Diagnosis of the unborn foetus is also possible during pregnancy. The care of people with sickle cell disease may include infection prevention with vaccination and antibiotics, high fluid intake, folic acid supplementation, and pain medication. Other measures may include blood transfusion and the medication hydroxycarbamide (hydroxyurea). In 2023, new gene therapies were approved involving the genetic modification and replacement of blood forming stem cells in the bone marrow. As of 2021, sickle cell disease is estimated to affect about 7.7 million people worldwide, directly causing an estimated 34,000 annual deaths and a contributory factor to a further 376,000 deaths. About 80% of sickle cell disease cases are believed to occur in sub-Saharan Africa. It also occurs to a lesser degree among people in parts of India, Southern Europe, West Asia, North Africa and among people of African origin (sub-Saharan) living in other parts of the world. The condition was first described in the medical literature by American physician James B. Herrick in 1910.
=== Glycine === Glycine is biosynthesized from serine, catalyzed by serine hydroxymethyltransferase (SHMT). The enzyme effectively replaces a hydroxymethyl group with a hydrogen atom. SHMT is coded by the gene glyA. The regulation of glyA is complex and is known to incorporate serine, glycine, methionine, purines, thymine, and folates, The full mechanism has yet to be elucidated. The methionine gene product MetR and the methionine intermediate homocysteine are known to positively regulate glyA. Homocysteine is a coactivator of glyA and must act in concert with MetR. On the other hand, PurR, a protein which plays a role in purine synthesis and S-adeno-sylmethionine are known to down regulate glyA. PurR binds directly to the control region of glyA and effectively turns the gene off so that glycine will not be produced by the bacterium.
== Types of standard solutions == Standard solutions can be categorized by the type of analyte used to prepare them. These analytical standards can either be a primary standard or a secondary standard.
The Canadas were the main front in the War of 1812 between the United States and the United Kingdom. Peace came in 1815; no boundaries were changed. Immigration resumed at a higher level, with over 960,000 arrivals from Britain between 1815 and 1850. New arrivals included refugees escaping the Great Irish Famine as well as Gaelic-speaking Scots displaced by the Highland Clearances. Infectious diseases killed between 25 and 33 percent of Europeans who immigrated to Canada before 1891. The desire for responsible government resulted in the abortive Rebellions of 1837. The Durham Report subsequently recommended responsible government and the assimilation of French Canadians into English culture. The Act of Union 1840 merged the Canadas into a united Province of Canada and responsible government was established for all provinces of British North America east of Lake Superior by 1855. The signing of the Oregon Treaty by Britain and the United States in 1846 ended the Oregon boundary dispute, extending the border westward along the 49th parallel. This paved the way for British colonies on Vancouver Island (1849) and in British Columbia (1858). The Anglo-Russian Treaty of Saint Petersburg (1825) established the border along the Pacific coast, but, even after the US Alaska Purchase of 1867, disputes continued about the exact demarcation of the Alaska–Yukon and Alaska–British Columbia border.

To supply the markets of Southern China, Makassarese trepangers traded with the Aboriginal Australians of Arnhem Land from at least the 18th century and likely considerably earlier. This Makassan contact with Australia is the first recorded example of interaction between the inhabitants of the Australian continent and their Asian neighbours. This contact had a major impact on the Indigenous Australians. The Makassarese exchanged goods such as cloth, tobacco, knives, rice and alcohol for the right to trepang coastal waters and employ local labour. Makassar pidgin became a lingua franca along the north coast among different Indigenous Australian groups who were brought into greater contact with each other by the seafaring Makassan culture. Archeological remains of Makassan contact, including trepang processing plants from the 18th and 19th centuries, are still found at Australian locations such as Port Essington and Groote Eylandt, and the Makassar-planted tamarind trees (native to Madagascar and East Africa).
The McDLT was served in a specialized, extra-wide dual-compartment polystyrene (Styrofoam) container. One side held the bottom bun and the hot beef patty, while the other side held the top bun, lettuce, tomato, American cheese, pickles, and sauces. The customer was instructed to flip the "cool" side onto the "hot" side immediately before eating. The dual-compartment clamshell packaging created operational challenges in McDonald's kitchens. The container was nearly double the size of a standard burger box, which required special equipment for storing it after preparation to heat one side while cooling the other, and it occupied twice as much space in the bins (chutes) behind the counter. Critics also noted that the empty space above the "hot" side allowed the beef patty to quickly lose heat, often resulting in a tepid sandwich by the time it was eaten. Serious Eats later pointed to uneven temperature control and the need for customers to assemble the sandwich as possible reasons for its failure.
=== Structure of graphite and its intercalation compounds === In 1859, Benjamin Brodie noted the highly lamellar structure of thermally reduced graphite oxide. Researchers used X-ray crystallography in an attempt to determine the structure of graphite. The lack of large single crystal graphite specimens contributed to the independent development of X-ray powder diffraction by Peter Debye and Paul Scherrer in 1915, and Albert Hull in 1916. However, neither of their proposed structures was correct. In 1918, Volkmar Kohlschütter and P. Haenni described the properties of graphite oxide paper. The structure of graphite was successfully determined from single-crystal X-ray diffraction by J. D. Bernal in 1924, while subsequent research tweaked the unit cell parameters. The theory of graphene was first explored by P. R. Wallace in 1947 as a starting point for understanding the electronic properties of 3D graphite. The emergent massless Dirac equation was separately pointed out in 1984 by Gordon Walter Semenoff, and by David P. Vincenzo and Eugene J. Mele. Semenoff emphasized the occurrence in a magnetic field of an electronic Landau level precisely at the Dirac point. This level is responsible for the anomalous integer Quantum Hall effect.
== Early life and education == Wittliff was born on June 15, 1938 in Taft, Texas, of primarily Eastern European heritage. He is a 6th generation Texan and direct descendant of John Cryer (Crier), who was recruited to settle Texas as part of Stephen F. Austin's original 300 families. He, his brother Bill and their mother Laura moved to Blanco when they were boys. In 1956, Wittliff graduated from San Marcos Academy, a college preparatory school that emphasized military discipline and training at the time. After 2 years of studying Chemical Engineering at Texas Tech University where he was a writer for The Daily Toreador and the National Publication Headquarters for the Arnold Air Society, he received a bachelor's degree in chemistry from University of Texas at Austin. While at UT, Wittliff worked at Clyde Campbell University Shop and modeled to support his education. Wittliff then earned an M. S. Degree in Biochemistry at Louisiana State University, School of Medicine. Wittliff's family moved to the University of Texas at Austin where he was awarded an National Defense Education Act (NDEA) Fellowship, a program influenced by the launch of the Sputnik satellite by the Soviets. Wittliff received his Ph.D. degree at The University of Texas at Austin in 1967. He then received an NIH Postdoctoral Award to study in the Laboratory of Professor Francis T. Kenney in the Biology Division at Oak Ridge National Laboratory.

== Mechanism of action == Like morphine, pethidine exerts its analgesic effects by acting as an agonist at the μ-opioid receptor. Pethidine is often employed in the treatment of postanesthetic shivering. The pharmacologic mechanism of this antishivering effect is not fully understood, but it may involve the stimulation of κ-opioid receptors. Pethidine has structural similarities to atropine and other tropane alkaloids and may have some of their effects and side effects. In addition to these opioidergic and anticholinergic effects, it has local anesthetic activity related to its interactions with sodium ion channels. Pethidine's apparent in vitro efficacy as an antispasmodic agent is due to its local anesthetic effects. It does not have antispasmodic effects in vivo. Pethidine also has stimulant effects mediated by its inhibition of the dopamine transporter (DAT) and norepinephrine transporter (NET). Pethidine will substitute for cocaine in animals trained to discriminate cocaine from saline, probably as a result of its inhibitory actions on DAT and NET. Several analogs of pethidine such as 4-fluoropethidine have been synthesized that are potent inhibitors of the reuptake of the monoamine neurotransmitters dopamine and norepinephrine via DAT and NET. It has also been associated with cases of serotonin syndrome, suggesting some interaction with serotonergic neurons, but the relationship has not been definitively demonstrated. It is more lipid-soluble than morphine, resulting in a faster onset of action.
=== Fluorescence of recombinant resilin === One unique property of rec1-Resilin is its ability to be identified due to autofluorescence. Fluorescence for resilin stems primarily from dityrosine, which are the result of crosslinks of tyrosine residues. When ultraviolet light irradiates a sample of rec1-Resilin at 315 nm to 409 nm emissions, the rec1-Resilin begins to show blue fluorescence. An example of the blue fluorescence exhibited by the dityrosine residues in resilin is shown in the figure below of a flea.
The two substrates of this enzyme are L-arogenic acid (shown as its conjugate base arogenate) and oxidised nicotinamide adenine dinucleotide (NAD+). Its products are L-tyrosine, reduced NADH, and carbon dioxide. This enzyme belongs to the family of oxidoreductases, specifically those acting on the CH-CH group of donor with NAD+ or NADP+ as acceptor. The systematic name of this enzyme class is L-arogenate:NAD+ oxidoreductase (decarboxylating). Other names in common use include arogenic dehydrogenase (ambiguous), cyclohexadienyl dehydrogenase, pretyrosine dehydrogenase (ambiguous), and L-arogenate:NAD+ oxidoreductase. This enzyme participates in phenylalanine, tyrosine and tryptophan biosynthesis and novobiocin biosynthesis.

It is a synthetic peptide built from 44 amino acids arranged in the same order as human growth hormone-releasing hormone. A short fatty-acid chain, described as a trans-3-hexenoyl group, is attached to the first amino acid. The finished molecule is formulated as a sterile powder that is dissolved before use.
No. It is a releasing-factor analog that signals the pituitary gland to secrete growth hormone, whereas recombinant growth hormone is the hormone itself administered directly. The two are chemically distinct and act at different points in the same endocrine pathway. This distinction is often lost in informal discussion.
Native growth hormone-releasing hormone is broken down within minutes by dipeptidyl peptidase-4 in the bloodstream. Adding the hexenoyl group at the N-terminus shields the peptide from that enzyme. The modification does not change the receptor it targets, only how long the peptide survives in circulation.
It mirrors the 44-residue form of human growth hormone-releasing hormone. A hexenoyl group on the N-terminal tyrosine distinguishes it from the unmodified hormone. The change is intended to improve resistance to enzymatic breakdown.
