This is a working overview of TH9507, written for readers who want more than a one-paragraph summary but less than a textbook.

Measured responses usually involve growth hormone and insulin-like growth factor 1, known as IGF-1. Growth hormone rises in bursts and is difficult to sample reliably, while IGF-1 shifts more slowly and can be assessed from a single blood draw. Studies therefore treat IGF-1 as the more practical pharmacodynamic marker. Both are indirect, showing that the receptor was engaged rather than that the peptide reached a particular concentration. Direct exposure measurement requires an assay aimed at the molecule itself.
Published work tends to frame tesamorelin as a tool for studying the GHRH axis and as a compound with measurable effects on body composition. Reports often describe visceral adipose tissue as an endpoint, assessed by imaging rather than by inference. Analytical sections commonly describe liquid chromatography with tandem mass spectrometry to confirm identity and purity, because immunoassays may cross-react with related fragments. Where results diverge between studies, differences in assay choice, sampling timing, and population are frequent explanations offered. Whether effects persist after treatment stops remains an open question.
Tesamorelin occupies a narrow position among agents that act on the growth hormone axis. Unlike growth hormone itself, which is given as replacement, it stimulates the pituitary to release the hormone in pulses, so the downstream increase in insulin-like growth factor 1 depends on intact somatotroph function. Other peptides in the same family include shorter GHRH fragments and synthetic secretagogues with different stability profiles. Several points remain unresolved, including whether the reduction in visceral fat translates into fewer cardiovascular events, what happens to metabolic markers after long-term use, and how the drug compares with lifestyle or surgical approaches.
Tesamorelin is a synthetic peptide that belongs to the growth hormone-releasing hormone family and contains the same forty-four amino acid sequence as endogenous GHRH, extended at the amino terminus by a trans-3-hexenoyl group. That small fatty acid modification protects the peptide from rapid cleavage by dipeptidyl peptidase-4, the enzyme that shortens the half-life of native GHRH to only a few minutes. Chemically the compound is produced by solid-phase peptide synthesis, purified by chromatography, and supplied as a sterile lyophilized powder for reconstitution.

Development work on the compound, originally designated TH9507, focused on conditions in which reduced growth hormone signaling is thought to contribute to altered body composition. The United States Food and Drug Administration approved it in 2010 for the treatment of excess visceral abdominal fat in adults with human immunodeficiency virus infection and lipodystrophy. Later research examined other populations, including adults with mild cognitive impairment, where a large trial did not meet its primary endpoints. This mixed record illustrates how a single mechanism can produce clear effects in one setting and inconclusive results in another.
Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
Measuring the effect of a growth hormone-releasing hormone analogue requires markers that reflect pituitary output rather than the peptide itself. The two most frequently used are growth hormone and insulin-like growth factor 1. Growth hormone fluctuates sharply across the day and responds to sleep, stress, and meals, so isolated readings can be difficult to interpret. Insulin-like growth factor 1 changes more slowly and is often treated as the more stable integrated marker of axis activity.
Because growth hormone is released in pulses, single measurements can misrepresent overall secretion. Investigators sometimes use repeated sampling or overnight profiles to capture the pattern rather than a single value. Provocative testing, in which a stimulus is given and the response is tracked over time, offers another way to characterize the axis. Each approach carries trade-offs between sensitivity, burden on the participant, and the influence of non-target variables.
In February 1917, revolution broke out in Russia in which workers, soldiers and peasants established soviets, the monarchy was forced into exile fell and a provisional government was formed until the election of a constituent assembly. Alexander Kerensky, a Russian lawyer and revolutionary, became a key political figure in the Russian Revolution of 1917. After the February Revolution, Kerensky joined the newly formed Russian Provisional Government, first as Minister of Justice, then as Minister of War and after July as the government's second Minister-Chairman. A leader of the moderate socialist Trudovik faction of the Socialist Revolutionary Party known as the Labour Group, Kerensky was also the vice-chairman of the powerful Petrograd Soviet. After failing to sign a peace treaty with the German Empire to exit from World War I which led to massive popular unrest against the government cabinet, Kerensky's government was overthrown on 7 November by the Bolsheviks led by Vladimir Lenin in the October Revolution. Soon after the October Revolution, the Russian Constituent Assembly elected Socialist-Revolutionary leader Victor Chernov as President of a Russian Republic, but it rejected the Bolshevik proposal that endorsed the Soviet decrees on land, peace and workers' control and acknowledged the power of the Soviets of Workers', Soldiers' and Peasants' Deputies.
== Role in wound healing == Wounds to the skin will be repaired in part by the migration of keratinocytes to fill in the gap created by the wound. The first set of keratinocytes to participate in that repair come from the bulge region of the hair follicle and will only survive transiently. Within the healed epidermis they will be replaced by keratinocytes originating from the epidermis. At the opposite, epidermal keratinocytes, can contribute to de novo hair follicle formation during the healing of large wounds. Functional keratinocytes are needed for tympanic perforation healing. A 2026 phase I clinical trial involving five patients reported that allogeneic cultured keratinocyte sheets were feasible and caused no serious adverse events when used to treat deep second-degree burns.
=== Further medieval development === The Fisher King's next development occurred around the end of the 12th century in Robert de Boron's Joseph d'Arimathie, the first work to connect the Grail with Jesus. Here, the "Rich Fisher" is called Bron, a name similar enough to Bran to suggest a relationship, and said to be the brother-in-law of Joseph of Arimathea, who had used the Grail to catch Christ's blood before laying him in the sepulchre. Joseph founds a religious community that travels eventually to Britain and entrusts the Grail to Bron (who is called the "Rich Fisher" because he catches a fish eaten at the Grail table). Bron founds the line of Grail keepers that eventually includes Perceval. The Lancelot-Grail (Vulgate) prose cycle includes a more elaborate history of the Fisher King. Many in his line are wounded for their failings, beginning with his ancestor Alain (the "Rich Fisherman"), and the only two that survived till the Grail Quest are the Maimed King, named Pellehan (Pellean, Pelleam, Pellehem), and the Fisher King, his son Pelles. The Vulgate Merlin Continuation further features besides his father, unnamed there, a brother of Pelles also named Alain, and in one variant also their brother and their cousin both named Pellinor, all featuring as Grail Kings of the Fisher and/or Maimed kind. Pelles engineers the birth of Galahad by tricking Lancelot into bed with his daughter Elaine, and it is prophesied that Galahad will achieve the Grail and heal the Wasteland and the Maimed King.
=== Endosymbionts === The Oceanospirillales symbionts are found in the specialized roots of all Osedax species, and play a major role in accelerating the degradation process of bones, as well as facilitating nutrient uptake for the Osedax. Oceanospirillales are known for their ability to degrade complex organic compounds. Campylobacterales are abundant along the trunk of the Osedax according to a 2023 study. Different genera in this order are found in Osedax at different points during the whale's degradation:
== Impact == Anderson is credited with the invention of the hamburger bun as well as "the kitchen as assembly line, and the cook as infinitely replaceable technician," hence giving rise to the modern fast-food phenomenon. Due to White Castle's innovation of having chain-wide standardized methods, customers could be sure that they would receive the same product and service in every White Castle restaurant. Ingram's business savvy was responsible not only for White Castle's success but also for the popularization of the hamburger. Time ranked the White Castle slider "The Most Influential Burger of All Time" in its January 14, 2014, edition. The restaurant is a central plot device of the comedy film Harold & Kumar Go to White Castle.

The US lost two nuclear submarines during this time: USS Thresher due to equipment failure during a test dive while at its operational limit, and USS Scorpion due to unknown causes. During the Indo-Pakistani War of 1971, the Pakistan Navy's Hangor sank the Indian frigate INS Khukri. This was the first sinking by a submarine since World War II. During the same war, Ghazi, a Tench-class submarine on loan to Pakistan from the US, was sunk by the Indian Navy. It was the first submarine combat loss since World War II. In 1982 during the Falklands War, the Argentine cruiser General Belgrano was sunk by the British submarine HMS Conqueror, the first sinking by a nuclear-powered submarine in war. Some weeks later, on 16 June, during the Lebanon War, an unnamed Israeli submarine torpedoed and sank the Lebanese coaster Transit, which was carrying 56 Palestinian refugees to Cyprus, in the belief that the vessel was evacuating anti-Israeli militias. The ship was hit by two torpedoes, managed to run aground but eventually sank. There were 25 dead, including her captain. The Israeli Navy disclosed the incident in November 2018.
Cell membranes are generally impermeable to charged or large, polar molecules because of the lipophilic fatty acyl chains comprising their interior. Many biologically important molecules, including a number of pharmaceutical agents, are organic weak acids that can cross the membrane in their protonated, uncharged form but not in their charged form (i.e., as the conjugate base). For this reason the activity of many drugs can be enhanced or inhibited by the use of antacids or acidic foods. The charged form, however, is often more soluble in blood and cytosol, both aqueous environments. When the extracellular environment is more acidic than the neutral pH within the cell, certain acids will exist in their neutral form and will be membrane soluble, allowing them to cross the phospholipid bilayer. Acids that lose a proton at the intracellular pH will exist in their soluble, charged form and are thus able to diffuse through the cytosol to their target. Ibuprofen, aspirin and penicillin are examples of drugs that are weak acids.
==== Hemisphere effect ==== The northern and southern hemispheres have atmospheric circulation systems that are sufficiently independent of each other that there is a noticeable time lag in mixing between the two. The atmospheric 14C/12C ratio is lower in the southern hemisphere, with an apparent additional age of about 40 years for radiocarbon results from the south as compared to the north. This is because the greater surface area of ocean in the southern hemisphere means that there is more carbon exchanged between the ocean and the atmosphere than in the north. Since the surface ocean is depleted in 14C because of the marine effect, 14C is removed from the southern atmosphere more quickly than in the north. The effect is strengthened by strong upwelling around Antarctica.
== Background == Bhagwat Singh Mewar was a patron of numerous trusts throughout his lifetime. On 20 October 1969, he founded the MMCF by providing a substantial endowment and donating key sections of the City Palace in Udaipur for its establishment. In his will and testament of 1984, he reconstituted the institution of the Maharana for the MMCF, so that the office of Maharana shall continue in perpetuity. After his death, his son Arvind Singh Mewar was appointed Chairman and Managing Trustee of the MMCF. Following the death of Arvind Singh Mewar on 16 March 2025, his son, Lakshyaraj Singh Mewar, now functions as the Managing Trustee of the trust.
==== Sirenia research ==== Saha, Bhardwaj & Bajpai (2026) study the restructuring of the sirenian skull throughout the evolutionary history of the group on the basis of data from extant and fossil taxa. Wagner et al. (2026) provide a methodology for quantification of the degree of pachyostosis in ribs and vertebrae of sirenians on the basis of the study of extant and extinct taxa. Pérez-Ramos et al. (2026) present a virtual reconstruction of the vertebral column of Pezosiren portelli. New fossil material of sirenians with dugongid affinities, associated with an invertebrate assemblage that might be indicative of presence of seagrass habitats, is described from the Oligocene strata of the Maniyara Fort Formation (India) by Joshi et al. (2026). Shinmura & Sawada (2026) reconstruct diets of sirenians from Miocene to Pliocene marine sediments in Hokkaido (Japan) on the basis of stable carbon isotope ratios of fatty acids and sterols extracted from their bones, providing evidence of Metaxytherium feeding on seagrass and evidence of feeding of Dusisiren and the Takikawa sea cow on seaweed.

It acts on the growth hormone–releasing hormone receptor, a Gs-coupled receptor found on pituitary somatotroph cells. Activation raises cAMP and prompts pulsatile hormone release.
IGF-1 reflects growth hormone activity but changes slowly and can be measured from one sample. Growth hormone itself is pulsatile, which makes single measurements hard to interpret.
The receptor pathway is well described, but how individual responses vary and what governs long-term outcomes remain open questions. Reported differences across studies are often attributed to assay and population factors.
It is a laboratory-made peptide of forty-four amino acids whose sequence matches human growth hormone-releasing hormone, with a modified amino terminus. The modification is a short unsaturated fatty acid chain attached to the first residue. This change slows enzymatic breakdown and lengthens the time the peptide stays active in circulation.
